Overview

A combination therapy of afatinib, an irreversible ErbB family blocker, and prednisone, a corticosteroid. Afatinib is a potent and selective, irreversible ErbB family blocker that covalently binds to and blocks signaling from all homo and heterodimers formed by the ErbB family members EGFR (ErbB1), HER2 (ErbB2), ErbB3, and ErbB4. It specifically targets mutations in the epidermal growth factor receptor (EGFR), making it effective for certain types of lung cancer. Prednisone is a corticosteroid that helps reduce inflammation and manage immune reactions. When combined with afatinib, it may help manage treatment-related adverse events while potentially enhancing therapeutic efficacy. This combination is being investigated for advanced squamous non-small cell lung cancer and has shown potential benefit in managing EGFR-TKI-induced interstitial lung disease.
