Overview

This is a four-drug combination regimen consisting of afatinib, ribavirin, carboplatin, and paclitaxel. Afatinib is an irreversible inhibitor of the ErbB family of tyrosine kinases (including EGFR/HER1 and HER2), used primarily in non-small cell lung cancer. Ribavirin is a nucleoside analog antiviral that inhibits viral RNA synthesis and mRNA capping, mainly used for hepatitis C and some viral hemorrhagic fevers. Carboplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis, commonly used in various solid tumors. Paclitaxel stabilizes microtubules to inhibit cell division, widely used in cancers such as ovarian, breast, and lung cancer. This specific combination has been studied as induction chemotherapy for HPV-associated locally advanced oropharyngeal squamous cell carcinoma (SCC), with phase I studies showing it to be safe and well tolerated[1][2][3]. The regimen aims to leverage the complementary mechanisms of action for enhanced antitumor activity.
