A combination therapy consisting of afatinib, a pan-ErbB tyrosine kinase inhibitor, and sotorasib (LUMAKRAS), a KRAS G12C inhibitor. This combination targets both KRAS G12C mutations and the ErbB family of receptors, potentially overcoming resistance mechanisms to KRAS inhibition alone. The combination has shown antitumor activity in patients with KRAS G12C-mutated non-small cell lung cancer (NSCLC), including those who previously progressed on sotorasib monotherapy.
