Overview

Afelimomab is a F(ab')2 fragment of a murine monoclonal antibody targeting human tumor necrosis factor alpha (TNFα). It was developed as an anti-TNFα therapy for the treatment of sepsis and septic shock. Afelimomab works by binding to TNFα, thereby inhibiting its pro-inflammatory activity, which leads to reduced levels of interleukin-6 and attenuation of the inflammatory response in severe sepsis. Clinical trials showed that afelimomab was safe, biologically active, and well tolerated in patients with severe sepsis, resulting in modest reductions in 28-day all-cause mortality and organ dysfunction severity among patients with elevated interleukin-6 levels. The drug was originally developed by Abbott Laboratories (now part of AbbVie) but development has been discontinued for all indications[1][2][3][4][5].
