Drug intelligence / Profile preview
Affimer A68 is a preclinical research compound designed as an Affimer, a small (~12 kDa) engineered non-antibody binding protein scaffold based on the cystatin fold. Developed at the University of Leeds, Affimer A68 was selected from a phage display library to bind and inhibit alpha-2-antiplasmin (plasmin inhibitor, PI), the primary physiological inhibitor of fibrinolysis. By blocking the action of alpha-2-antiplasmin, Affimer A68 enhances fibrin clot lysis and reduces clot lysis times in purified systems, plasma, and whole blood. This targeted approach aims to modulate the hypofibrinolytic environment, particularly in conditions like diabetes, to reduce the risk of thrombosis and vascular occlusion without increasing bleeding risks.
