Drug intelligence / Profile preview
Affimer RV8 is an engineered non-antibody scaffold protein (Affimer) designed to specifically bind and inhibit plasminogen activator inhibitor-1 (PAI-1). Developed by researchers at the University of Leeds, Affimer RV8 was identified from a phage display library. PAI-1 is a key physiological inhibitor of fibrinolysis that is frequently elevated in patients with type 2 diabetes, contributing to a prothrombotic state and impaired clot breakdown (hypofibrinolysis). By selectively inhibiting PAI-1 activity, Affimer RV8 facilitates the conversion of plasminogen to plasmin, thereby enhancing fibrin clot lysis without disrupting the physiological structure of the fibrin network. This therapeutic approach is being investigated as a novel strategy to reduce thrombosis risk and improve fibrinolytic balance in patients with diabetes and other cardiovascular diseases.
