Overview

Affinity-enhanced NY-ESO-1 TCR T-cell therapy is an adoptive cell therapy modality in which a patient's autologous T cells are genetically engineered to express a high-affinity T-cell receptor (TCR) specific for the NY-ESO-1 (CTAG1B) cancer-testis antigen. Naturally occurring TCRs against tumor antigens often have low affinity due to central tolerance; these engineered TCRs are modified in their complementarity-determining regions (CDRs) to enhance binding avidity to the NY-ESO-1 peptide presented by HLA-A*02. This approach allows the T cells to effectively recognize and kill tumor cells expressing NY-ESO-1. Key developers include Adaptimmune Therapeutics (afamitresgene autoleucel) and GSK (letetresgene autoleucel). The therapy has shown significant efficacy in solid tumors like synovial sarcoma and myxoid/round cell liposarcoma, as well as hematologic malignancies like multiple myeloma.
