Overview

AFG3L2 overexpression adenovirus is a viral vector-based gene therapy tool designed to increase the expression of the AFG3L2 (AFG3-like protein 2) gene. AFG3L2 is a critical component of the m-AAA protease complex located in the inner mitochondrial membrane, where it plays a vital role in mitochondrial protein quality control and the assembly of the oxidative phosphorylation system. In preclinical models of inflammatory bowel disease, specifically Crohn's disease, the overexpression of AFG3L2 has been shown to inhibit ferroptosis in intestinal epithelial cells. This protective effect is mediated through the activation of the peroxisome proliferator-activated receptor-alpha (PPARA) signaling pathway, which subsequently upregulates the transcription of glutathione peroxidase 4 (GPX4), a key enzyme that prevents lipid peroxidation. By maintaining mitochondrial ultrastructure and reducing reactive oxygen species (ROS) production, this gene therapy approach aims to suppress intestinal inflammation and preserve mucosal integrity.
