Afimkibart is an investigational fully human immunoglobulin G1 monoclonal antibody that neutralizes tumor necrosis factor-like ligand 1A, preventing TL1A signaling through death receptor 3 and thereby modulating inflammatory and fibrotic pathways implicated in immune-mediated disease. Originally developed by Pfizer as PF-06480605 and subsequently known as RVT-3101, it was acquired by Roche through the Telavant transaction and is being developed principally for moderately to severely active ulcerative colitis and Crohn's disease, with additional clinical development in atopic dermatitis and rheumatoid arthritis. Roche-sponsored Phase III studies are ongoing in ulcerative colitis, including a pediatric induction and maintenance study.
