Aflibercept + docetaxel is a combination therapy consisting of aflibercept, a recombinant human fusion protein that acts as a decoy receptor for vascular endothelial growth factor (VEGF)-A, VEGF-B, and placental growth factor (PlGF), and docetaxel, a cytotoxic taxane chemotherapy agent. Aflibercept inhibits angiogenesis by binding to VEGF-A, VEGF-B, and PlGF with high affinity, preventing their interaction with native receptors on the surface of endothelial cells. Docetaxel promotes microtubule assembly while inhibiting disassembly, leading to cell cycle arrest and apoptosis in rapidly dividing tumor cells. This combination has been investigated primarily in advanced solid tumors such as metastatic castrate-resistant prostate cancer and other advanced malignancies[1][2][4][5].
