Aflibercept + oxaliplatin refers to the use of both aflibercept, a recombinant fusion protein that acts as a decoy receptor for vascular endothelial growth factor (VEGF), and oxaliplatin, a platinum-based chemotherapy agent. Aflibercept inhibits angiogenesis by binding VEGF-A, VEGF-B, and placental growth factor (PlGF), preventing their interaction with endogenous receptors and thus reducing tumor blood vessel formation. Oxaliplatin is a cytotoxic small molecule that forms DNA crosslinks, inhibiting DNA replication and transcription in rapidly dividing cells. While aflibercept is approved for use in combination with FOLFIRI (fluorouracil/leucovorin/irinotecan) after progression on an oxaliplatin-containing regimen in metastatic colorectal cancer, there is no evidence supporting or describing the clinical use of aflibercept directly combined with oxaliplatin as a standard regimen[1][2][3][4][5]. Both drugs are used sequentially or in different combinations for metastatic colorectal cancer.
