Overview

AFM11 is a bispecific, tetravalent humanized recombinant antibody construct (TandAb) developed by Affimed Therapeutics for the treatment of hematologic malignancies, specifically relapsed or refractory non-Hodgkin lymphoma (NHL) and acute lymphoblastic leukemia (ALL)[1][4][6]. It is designed to bind simultaneously to CD19 on B-cells and CD3 on T-cells. By bridging these two cell types, AFM11 recruits and activates T-cells to induce cytotoxicity against malignant B-cells through antibody-dependent cell-mediated cytotoxicity. The drug does not directly kill cancer cells but stimulates the immune system’s T-cell response to target and destroy CD19-positive cancer cells[4][5][7]. Clinical development was discontinued after phase 1 trials due to safety concerns including severe neurological adverse events[1][6].
