AFM13 + cord blood-derived natural killer cells is an allogeneic cell-based immunotherapy developed by M.D. Anderson Cancer Center in collaboration with Affimed. The therapy consists of umbilical cord blood-derived natural killer (NK) cells that are activated with a cytokine cocktail (IL-12, IL-15, and IL-18), expanded using artificial antigen-presenting cells, and subsequently pre-complexed with AFM13 (acimtamig). AFM13 is a bispecific tetravalent Innate Cell Engager (ICE) that binds to CD16A on NK cells and CD30 on lymphoma cells, creating a bridge that enables the NK cells to target and destroy CD30-positive cancer cells. This approach creates CAR-like responses without the need for viral-mediated genetic modification of the cells. The therapy is primarily investigated for heavily pretreated patients with relapsed or refractory CD30-positive malignancies, including Hodgkin lymphoma and various types of non-Hodgkin lymphoma.
