AFM21

AFM21 is a preclinical, tetravalent bispecific TandAb antibody being developed by Affimed to redirect T cells against tumors expressing the mutant epidermal growth factor receptor variant III (EGFRvIII), a tumor-specific alteration frequently found in glioblastoma and other solid tumors. Designed with Affimed’s TandAb platform, AFM21 incorporates binding domains for CD3 on T cells and EGFRvIII on tumor cells, enabling formation of an immunological synapse that promotes targeted T-cell–mediated cytotoxicity while sparing normal EGFR-wild-type tissues.[2][5][11] Affimed has reported that AFM21, together with a related NK-cell engager AFM22, demonstrates potent and selective in vitro cytotoxicity against EGFRvIII-positive tumor cells, and the company has indicated plans for IND-enabling studies as part of its solid tumor immunotherapy pipeline.[2][5]
