AFM24 + SNK01 is an investigational combination therapy consisting of two components: **AFM24**, a tetravalent bispecific IgG1-scFv fusion monoclonal antibody that simultaneously targets epidermal growth factor receptor (EGFR) on tumor cells and CD16A (FcγRIIIA) on natural killer (NK) cells, thereby facilitating NK cell-mediated, antibody-dependent cellular cytotoxicity (ADCC) specifically against EGFR-expressing solid tumor cells; and **SNK01**, an autologous adoptive cell therapy composed of ex vivo expanded and activated NK cells with high CD16 expression, produced from patient-derived peripheral blood. AFM24 is designed to redirect highly activated NK cells (from SNK01) to EGFR-positive tumor cells, potentially overcoming resistance seen with traditional EGFR inhibitors and enhancing antitumor immune responses. The combination is under clinical evaluation for patients with advanced or metastatic EGFR-expressing solid tumors who are refractory to standard therapy. Developed by Affimed (AFM24) and NKGen Biotech (SNK01), the combination has shown promising disease control and a favorable safety profile in early-phase trials, with most adverse events being mild infusion reactions. The combination is administered intravenously, with both agents given once weekly in 28-day cycles[1][2][3][4][5][7].
