AFM26 is a bispecific, tetravalent Innate Cell Engager (ICE) developed by Affimed that simultaneously targets the tumor-associated antigen epithelial cell adhesion molecule (EpCAM) and the activating receptor CD16A (FcγRIIIA) on natural killer (NK) cells and macrophages. Built on the proprietary ROCK (Redirected Optimized Cell Killing) platform, AFM26 is designed to recruit and activate the innate immune system to destroy EpCAM-positive tumor cells. It binds with high affinity to both the 158V and 158F allotypes of CD16A, ensuring broad patient applicability. AFM26 is being explored as a monotherapy and as part of a combination strategy with adoptive NK cell transfers, offering an off-the-shelf approach for treating various EpCAM-expressing solid tumors while potentially overcoming the limitations of conventional monoclonal antibodies, such as competition with circulating IgG.
