Overview

AFM28 is a high-affinity bispecific, tetravalent antibody-based construct designed to engage natural killer (NK) cells for the treatment of CD123-expressing hematological malignancies, primarily acute myeloid leukemia (AML). It works by binding to CD16A on NK cells and CD123 on leukemic cells, redirecting NK cell cytotoxicity toward malignant blasts and leukemic stem/progenitor cells. This mechanism enables targeted elimination of AML cells while sparing healthy tissue. Developed by Affimed, AFM28 has demonstrated preliminary clinical efficacy and a manageable safety profile in phase 1 studies involving relapsed/refractory AML patients. The drug is also being explored for potential use in combination with allogeneic NK cell therapy and may have future applications in other CD123-positive malignancies[1][2][3][4][5].
