Drug intelligence / Profile preview
aFn-uPA (anti-fibrin urokinase plasminogen activator) is a preclinical-stage recombinant fusion protein designed as a targeted thrombolytic therapy. Developed by Synapse Research Institute and its spinout Mithridate, the construct consists of a urokinase-type plasminogen activator (uPA) genetically fused to a fibrin-specific single-domain antibody (sdAb/nanobody). By targeting fibrin directly without cross-reacting with fibrinogen, aFn-uPA localizes plasminogen activation to the site of a blood clot, thereby enhancing thrombolytic efficacy and reducing systemic bleeding risks compared to standard recombinant tissue-type plasminogen activators (rtPA) like alteplase. It is being investigated for acute thrombotic events, including ischemic stroke, ST-elevation myocardial infarction (STEMI), and pulmonary embolism.
