AFN18 is a personalized tumor neoantigen mRNA vaccine designed for the treatment of solid tumors. This investigational therapy utilizes next-generation sequencing to identify patient-specific tumor neoantigens—unique peptides arising from somatic mutations in the patient's cancer cells. The identified neoantigens are then encoded into an mRNA construct, which is formulated for administration (typically via lipid nanoparticles). Upon injection, the mRNA is taken up by antigen-presenting cells, leading to in situ production and presentation of these neoantigen peptides on MHC molecules. This process stimulates a robust and specific T cell-mediated immune response against tumor cells expressing those antigens, aiming to achieve targeted anti-tumor activity while minimizing off-target effects. The primary goal of AFN18 is to enhance anti-tumor immunity in patients with various types of solid tumors through individualized immunotherapy[1][2][8].
