Overview

AFN50 is an *in vivo* Chimeric Antigen Receptor (CAR) T-cell therapy developed by AlphaNa Bioscience for the treatment of autoimmune diseases, specifically systemic lupus erythematosus (SLE). The therapy utilizes a novel T-cell-targeted lipid nanoparticle (T-LNP) delivery system to encapsulate mRNA encoding a CAR specific for the B-lymphocyte antigen CD19. Unlike traditional *ex vivo* CAR-T therapies that require complex cell manufacturing outside the patient's body, AFN50 is designed for direct intravenous administration. Once in the bloodstream, the T-LNPs selectively deliver the mRNA payload to the patient's endogenous T cells, which then transiently express the CAR to identify and deplete pathogenic CD19-positive B cells. This approach aims to provide the therapeutic benefits of B-cell depletion while avoiding the need for lymphodepleting chemotherapy and the logistical challenges of *ex vivo* cell engineering.
