Overview

AFNT-111 is an investigational autologous T-cell receptor (TCR)-engineered cell therapy developed by Affini-T Therapeutics for the treatment of solid tumors harboring the KRAS G12V mutation. The therapy consists of patient-derived CD4+ and CD8+ T cells that are genetically modified to express a high-affinity, naturally occurring TCR specific for the KRAS G12V mutant peptide presented by HLA-A*11:01. This engineered receptor enables targeted recognition and killing of tumor cells expressing this oncogenic driver mutation. Preclinical studies have demonstrated potent and specific cytotoxicity against KRAS G12V-expressing tumor cells in models of breast, pancreatic, colon, lung, and ovarian cancers. The product is currently being evaluated in Phase 1 clinical trials for metastatic solid tumors including colorectal cancer, pancreatic adenocarcinoma, and non-small cell lung cancer[1][2][3][4].
