Overview

AFPTK1 is a gene therapy construct comprising the herpes thymidine kinase (HSVtk) gene under the transcriptional control of the alpha-fetoprotein (AFP) promoter. It is designed for suicide gene therapy, specifically targeting AFP-producing hepatocellular carcinoma. When delivered into cancer cells, the AFP promoter ensures selective expression of the HSVtk gene in cells that produce AFP. The expressed HSVtk enzyme then converts the co-administered prodrug ganciclovir (GCV) into a cytotoxic metabolite, which leads to apoptosis and suppression of tumor growth. The construct is typically encapsulated within hemagglutinating virus of Japan (HVJ)-liposomes for delivery.
