Overview

**Afuresertib + paclitaxel** is an investigational combination therapy of a pan-AKT inhibitor (afuresertib) and a taxane chemotherapy (paclitaxel), mainly studied in patients with platinum-resistant or refractory ovarian cancer. Afuresertib inhibits AKT, a central kinase in the PI3K/AKT signaling pathway that promotes tumor cell survival and proliferation, thereby aiming to restore chemosensitivity. Paclitaxel is an antineoplastic agent that stabilizes microtubules and inhibits cell division. This combination has been tested in phase 1 and phase 2 clinical trials but has not demonstrated significant benefit in overall or progression-free survival over paclitaxel alone, except possibly among biomarker-selected subpopulations with high pAKT expression[2][3][5][6].
