AFV-2101 is an investigational COVID-19 vaccine candidate developed by Affinivax (now a subsidiary of GSK) using the proprietary Multiple Antigen Presenting System (MAPS) platform. The MAPS technology utilizes a high-affinity, non-covalent biotin-rhizavidin binding mechanism to couple protective antigens, such as polysaccharides and proteins, into a single macromolecular complex. For AFV-2101, the vaccine incorporates the SARS-CoV-2 spike protein receptor-binding domain (RBD) to induce a dual immune response. Unlike traditional conjugate vaccines that primarily stimulate B-cells, the MAPS platform is designed to elicit both robust neutralizing antibody titers and a strong T-cell response (specifically Th1 and Th17), which may provide broader and more durable protection against emerging viral variants. Following GSK's acquisition of Affinivax in 2022, the development of this specific COVID-19 candidate was deprioritized as the company focused on its advanced pneumococcal MAPS programs.
