AFX-2401 is a small molecule therapeutic candidate being developed by Afecta Pharmaceuticals for the treatment of Attention Deficit Hyperactivity Disorder (ADHD). It is a sustained-release (SR) formulation of atomoxetine, a selective norepinephrine reuptake inhibitor (SNRI). By inhibiting the norepinephrine transporter (NET), atomoxetine increases the synaptic concentration of norepinephrine, particularly in the prefrontal cortex, which is thought to mediate its therapeutic effects in ADHD. The sustained-release profile of AFX-2401 is designed to optimize the drug's pharmacokinetic properties, potentially offering improved tolerability and a more consistent therapeutic effect throughout the day compared to immediate-release formulations. As of 2020, AFX-2401 was reported to be in Phase 2 clinical development.
