Overview

AG-636 is an orally available small molecule inhibitor of dihydroorotate dehydrogenase (DHODH), a key enzyme in the de novo pyrimidine synthesis pathway. By inhibiting DHODH, AG-636 blocks the fourth enzymatic step in pyrimidine biosynthesis, leading to impaired DNA and RNA synthesis and resulting in antineoplastic activity. Preclinical studies have shown that AG-636 has potent growth-inhibitory effects on hematologic cancer cell lines, particularly those derived from lymphomas and acute myeloid leukemia (AML), while its effect on solid tumor cell lines is relatively poor. The drug was originally developed by Agios Pharmaceuticals and later acquired by Servier for further development. It has been investigated primarily for advanced lymphoma and AML[1][4][5][6].
