AG019 + teplizumab is an investigational combination therapy for type 1 diabetes. AG019 is an oral capsule containing genetically engineered Lactococcus lactis bacteria designed to deliver human proinsulin and human interleukin-10 directly to the gut mucosa. This approach aims to induce antigen-specific immune tolerance by promoting regulatory T cell responses and reducing autoimmune destruction of pancreatic beta cells. Teplizumab is a humanized monoclonal antibody targeting CD3 on T lymphocytes; it modulates the immune response by deactivating autoreactive T cells involved in beta cell destruction. The combination seeks to synergistically preserve endogenous insulin production in newly diagnosed or early-stage type 1 diabetes patients[2][3][4][7].
