AG1024 is a small molecule tyrphostin that acts as a selective inhibitor of the insulin-like growth factor 1 receptor (IGF-1R) tyrosine kinase. It works by inhibiting the autophosphorylation of IGF-1R, thereby blocking downstream signaling pathways such as PI3K/Akt and MAPK, which are critical for cell proliferation and survival. AG1024 has been extensively studied in preclinical research for its potential antitumor effects, particularly in cancers where IGF-1R is overexpressed or plays a key role in resistance to other therapies, such as renal cell carcinoma and breast cancer. While it also inhibits the insulin receptor (IR), it demonstrates a higher selectivity for IGF-1R.
