Overview

AG614 is a synthetic, neuroinflammation-responsive promoter designed for use in central nervous system (CNS) gene therapies. Developed by Annogen using their proprietary SuRE (Survey of Regulatory Elements) screening platform, AG614 was engineered to provide conditional transgene expression that is activated during pathological inflammatory states and silenced upon resolution. In preclinical studies using the experimental allergic encephalitis (EAE) mouse model, AG614 demonstrated robust and specific induction in the spinal cord in response to neuroinflammation. This regulatory element is intended to improve the safety and efficacy of gene therapies for neurodegenerative disorders, such as multiple sclerosis, by ensuring therapeutic proteins are only produced when and where they are needed.
