AG73 is a synthetic dodecapeptide with the sequence RKRLQVQLSIRT, derived from the globular G4 domain of the laminin α1 chain. It is a highly specific ligand for syndecans, a family of cell-surface heparan sulfate proteoglycans (HSPGs), including Syndecan-1, Syndecan-2, and Syndecan-4. In the field of biotechnology and advanced drug delivery, AG73 is primarily utilized as a targeting moiety to facilitate the selective uptake of therapeutic payloads, such as liposomes, nanoparticles, and gene therapy vectors, into cells that overexpress syndecans. This overexpression is characteristic of various malignancies and vascular endothelial cells during angiogenesis. Upon binding to the heparan sulfate chains of syndecans, the AG73-linked complex is internalized via receptor-mediated endocytosis, effectively delivering the therapeutic agent into the cytoplasm.
