Ag85A-DC vaccine is an experimental immunotherapeutic vaccine composed of dendritic cells (DCs) engineered or transfected with the gene encoding Ag85A, a major secreted antigen from *Mycobacterium tuberculosis*. This strategy aims to harness the antigen-presenting capabilities of DCs together with robust immunostimulatory properties of Ag85A to induce potent, specific immune responses. The vaccine stimulates dendritic cell maturation, upregulates expression of costimulatory and MHC-II molecules, and induces strong proliferation and activation of both CD4+ and CD8+ T-cells, notably promoting Th1-type responses with increased interferon-gamma (IFN-γ) and interleukin-2 (IL-2) production. In animal models, intratumoral or subcutaneous administration of Ag85A-DC vaccine enhances cytotoxic T lymphocyte (CTL) activity, leading to significantly inhibited tumor growth in cancers such as colorectal carcinoma and bladder cancer, and increases infiltration of T-cells into tumors. It is under preclinical investigation as a strategy for cancer immunotherapy and tuberculosis vaccination.
