**aGalCer-OVA** is an experimental polymeric nanoparticle cancer-vaccine formulation that co-delivers the glycolipid invariant natural killer T-cell adjuvant alpha-galactosylceramide and the model tumor antigen ovalbumin. In murine B16F10-OVA melanoma models, it activated invariant natural killer T cells, promoted tumor infiltration by invariant natural killer T cells, natural killer cells, and ovalbumin-specific CD8-positive T cells, and enhanced cytotoxic-effector production including granzyme B and perforin. It showed prophylactic and therapeutic antitumor activity in mice and was more effective when combined with anti-CTLA-4 plus anti-PD-1 checkpoint blockade. This is an academic, preclinical model immunotherapy rather than a marketed or clinical-stage product.
