AGEN1223 is a novel bispecific antibody developed by Agenus for the treatment of advanced solid tumors. It is designed to selectively deplete regulatory T cells (Tregs) within the tumor microenvironment by targeting two antigens co-expressed on tumor-infiltrating Tregs. This selective depletion enhances antitumor immune responses while sparing peripheral Tregs and effector T cells. The mechanism involves antibody-dependent cell-mediated cytotoxicity (ADCC) and modulation of regulatory T lymphocytes[1][2][5]. Clinical development reached Phase 1 before being discontinued for strategic business reasons[2][7].
