Overview

AGEN1571 is a fully human IgG4κ monoclonal antibody that acts as a high-affinity antagonist of ILT2 (also known as LILRB1), an inhibitory receptor expressed on various immune cells. By binding to and blocking ILT2, AGEN1571 prevents its interaction with ligands such as HLA-G and other MHC class I molecules, thereby relieving immune suppression in the tumor microenvironment. This blockade enhances both innate and adaptive anti-tumor immunity by promoting activation of NK cells, NKT cells, CD8+ T effector cells, and re-polarizing macrophages toward a pro-inflammatory phenotype. The drug is being developed primarily for the treatment of advanced solid tumors and is currently in Phase 1 clinical trials. It may be used alone or in combination with other immunotherapies such as botensilimab (anti-CTLA-4) or balstilimab (anti-PD-1). The developer is Agenus[5][7][6][2].
