Overview

AGEN1571 + botensilimab is an investigational combination therapy for advanced solid tumors, being developed by Agenus. AGEN1571 is a novel anti-ILT2 monoclonal antibody that modulates tumor-associated macrophages, T, NK, and NKT cells to promote anti-tumor immune activity, overcoming resistance to PD-1-directed therapies. Botensilimab (AGEN1181) is an Fc-enhanced anti-CTLA-4 monoclonal antibody engineered to stimulate both innate and adaptive immune responses by robustly engaging activating Fc receptors on key immune cells, thereby activating T cells, depleting tumor-infiltrating regulatory T cells, and priming the tumor microenvironment for effective antitumor immunity. The combination is evaluated primarily in advanced solid tumors, including refractory microsatellite-stable metastatic colorectal cancer, with agents targeting distinct immune checkpoints to enhance and broaden the response to immunotherapy[1][2][3].
