Overview

AGEN1721 is a first-in-class, Fc-enhanced bifunctional bispecific antibody developed by Agenus. It targets both fibroblast activation protein alpha (FAP) and transforming growth factor beta (TGF-β), two key mediators of immune suppression in the tumor microenvironment. By simultaneously modulating FAP and TGF-β, AGEN1721 remodels the tumor stroma, enhances T-cell infiltration, and overcomes cancer-associated fibroblast-mediated immune suppression. This dual-targeting approach is designed to convert immunologically "cold" tumors into more immune-responsive environments, potentially improving outcomes in cancers that are resistant to conventional immunotherapies. Preclinical data demonstrate its ability to enhance antitumor responses by dismantling barriers to immune cell infiltration[1][2][6][8].
