AGEN1777 (also known as BMS-986442) is a first-in-class, Fc-enhanced bispecific monoclonal antibody designed to target and inhibit both TIGIT (T cell immunoreceptor with Ig and ITIM domains) and CD96 antigen, two major inhibitory immune checkpoint receptors expressed on T cells and natural killer (NK) cells. By simultaneously blocking these checkpoints, AGEN1777 aims to restore effector cell function and enhance anti-tumor immune responses. The drug leverages Agenus’ proprietary Fc-engineering platform for improved immune activation compared to conventional TIGIT antibodies. It is being developed in partnership between Agenus and Bristol Myers Squibb for the treatment of multiple solid tumors, including non-small cell lung cancer, colon cancer, gastric cancer, breast cancer, colorectal cancer, gastroesophageal junction carcinomas, head and neck squamous cell carcinoma. Clinical development includes studies as monotherapy or in combination with PD-(L)1 inhibitors such as nivolumab[1][2][4][5][6][7].
