**AGEN1777 + programmed cell death protein 1 inhibitor** is an experimental combination therapy comprising AGEN1777, a first-in-class, Fc-enhanced bispecific monoclonal antibody targeting TIGIT and CD96, together with an inhibitor of programmed cell death protein 1 (PD-1). The combination is designed to block major immune inhibitory receptors on T cells and NK cells, aiming to improve anti-tumor immune responses in patients with advanced solid tumors, including colon cancer and non-small cell lung cancer. AGEN1777 uses mammalian retrocyte and phage display technologies, and preclinical studies suggest that the dual targeting of TIGIT and CD96 may overcome resistance to anti-PD-1 or anti-TIGIT monotherapies. This combination has been tested in early-phase oncology trials, primarily via intravenous administration[1][4][6][7].
