AGEN2373 is a conditionally-active, fully human immunoglobulin G1 (IgG1) agonistic monoclonal antibody that targets the costimulatory receptor CD137 (also known as 4-1BB or tumor necrosis factor receptor superfamily member 9, TNFRSF9). It is designed to stimulate T cells and natural killer (NK) cells to promote a durable memory immune response against cancer. The drug selectively binds to a unique epitope on CD137, aiming to enhance antitumor immunity while minimizing hepatotoxicity and off-target effects commonly associated with systemic CD137 activation. Its mechanism of action relies on Fc-dependent cross-linking for conditional activation, which restricts its activity primarily to the tumor microenvironment. Developed by Agenus, AGEN2373 has demonstrated objective responses and disease control in heavily pretreated patients with advanced solid tumors in early-phase clinical trials. It is also being evaluated in combination with other immunotherapies such as botensilimab for PD-(L)1 refractory melanoma[1][2][3][6].
