Overview

AGEN307 is an experimental bispecific monoclonal antibody developed by Agenus for the treatment of advanced solid tumors. It is designed to simultaneously target **CD137 (4-1BB)** and **Programmed Death-Ligand 1 (PD-L1)**. The molecule functions by providing a localized co-stimulatory signal to T-cells through CD137 activation, which is conditionally triggered upon binding to PD-L1 in the tumor microenvironment. This dual-targeting approach is intended to enhance the potency of the immune response against cancer cells while reducing the systemic inflammatory side effects typically associated with systemic CD137 agonism. Although it showed promise in preclinical models, AGEN307 is no longer listed in Agenus's active clinical development pipeline.
