AGG-523 is an investigational small molecule drug developed as a selective oral inhibitor of aggrecanase 1 (ADAMTS4) and aggrecanase 2 (ADAMTS5), which are zinc-dependent endopeptidases implicated in the degradation of aggrecan in articular cartilage. By inhibiting these enzymes, AGG-523 aims to prevent cartilage breakdown associated with osteoarthritis. Preclinical studies demonstrated chondroprotective effects in animal models of osteoarthritis without the side effects seen with broad-spectrum matrix metalloproteinase inhibitors. The drug entered Phase 1 clinical trials for osteoarthritis but development was suspended after early-stage studies[1][2][3][4][5][6].
