**Agg-CLNP** is a preclinical, locally administered immunomodulatory polymer-nanoparticle formulation developed for prevention of rheumatoid arthritis onset and mitigation of recurrent rheumatoid arthritis flares. It encapsulates calcitriol and is surface-conjugated to an immunodominant aggrecan peptide, enabling uptake by dendritic cells in lymph nodes proximal to arthritic joints. In human monocyte-derived dendritic cells and mouse rheumatoid arthritis models, Agg-CLNP reduced dendritic-cell activation and HLA class II expression, increased CTLA-4 expression, suppressed local pathogenic T-helper 17 cells, delayed arthritis onset in combination with abatacept, and reduced post-corticosteroid-withdrawal flare severity. It has been reported by investigators at University of California San Diego and Cedars-Sinai, with no clinical trial phase reported.
