Drug intelligence / Profile preview
AGI-004 is a once-daily, controlled-release transdermal patch formulation of mecamylamine, a non-selective nicotinic acetylcholine receptor antagonist and ganglion blocker. Developed by AGI Therapeutics, the drug was designed as a novel anti-diarrheal agent for the treatment of chemotherapy-induced diarrhea (CID) and diarrhea-predominant irritable bowel syndrome (d-IBS). By utilizing a transdermal delivery system, AGI-004 aims to provide sustained drug exposure while minimizing the peak-to-trough fluctuations associated with oral administration, thereby reducing traditional non-gastrointestinal side effects such as hypotension while maintaining therapeutic effects on gut motility. Phase II clinical trials demonstrated positive results in patients with functional diarrhea and CID, showing good tolerability and a reduction in diarrheal symptoms.
