AGI-010 is a bimodal modified-release oral formulation of the proton pump inhibitor (PPI) omeprazole, developed by AGI Therapeutics using its proprietary CHRONAB technology. It was specifically designed to address nocturnal acid breakthrough (NAB) in patients with gastro-esophageal reflux disease (GERD). The formulation consists of a combination of delayed and controlled-release beads that create a bimodal plasma profile, extending drug release over a 6-8 hour period to maintain acid suppression throughout the night. By targeting the H+/K+-ATPase enzyme in gastric parietal cells, AGI-010 aimed to provide superior control of night-time gastric acidity compared to standard immediate-release PPIs. Development was initially co-sponsored by Axcan Pharma, but AGI Therapeutics regained full rights in 2009 before the product was eventually discontinued.
