AGI-101H is an allogeneic, gene-modified, whole-cell therapeutic cancer vaccine developed by AGIRx Ltd. for the treatment of advanced melanoma. It consists of a 1:1 mixture of two human melanoma cell lines, Mich1 and Mich2, which have been retrovirally transduced to express Hyper-IL6 (H6)—a fusion protein of interleukin-6 (IL-6) and its soluble receptor (sIL-6R). This genetic modification serves as a molecular adjuvant and induces a melanoma stem cell (MSC)-like phenotype in the vaccine cells, characterized by high ALDH1A1 activity. Immunization with AGI-101H is designed to elicit both cellular and humoral immune responses against MSCs, modulating T cell signaling pathways (TNF-α, TGF-β, and IL2-STAT5) and upregulating BCL6 to maintain long-term antigen-specific T cell memory.
