Aglatimagene besadenovec + celecoxib is an investigational combination therapy for cancer, primarily studied in solid tumors. Aglatimagene besadenovec (CAN-2409, also known as ADV-TK) is a replication-defective adenoviral vector engineered to deliver the herpes simplex virus thymidine kinase (HSV-tk) gene into tumor cells. Once inside the cell, HSV-tk expression sensitizes tumor cells to antiviral prodrugs such as valacyclovir or ganciclovir, which are converted into toxic metabolites that induce immunogenic cell death and stimulate an anti-tumor immune response[1][2][3][5]. Celecoxib is a selective cyclooxygenase-2 (COX-2) inhibitor with anti-inflammatory and potential antineoplastic properties. The rationale for combining these agents is to enhance local cytotoxicity and modulate the tumor microenvironment through both direct oncolytic effects and immunomodulation.
