AGN 199170 is a novel muscarinic receptor agonist developed by Allergan for the potential treatment of glaucoma. It is characterized by its lack of activity at the M3 muscarinic receptor subtype, which is the primary mediator of ocular side effects such as miosis (pupil constriction) and lens accommodation associated with traditional muscarinic agonists like pilocarpine. In preclinical studies using monkey models of glaucoma, AGN 199170 demonstrated significant intraocular pressure (IOP) lowering effects. This suggests that muscarinic receptors other than M3 are involved in the regulation of IOP, providing a pathway for developing glaucoma therapies with an improved side-effect profile.
