Drug intelligence / Profile preview
An agonistic anti-BTLA (B and T lymphocyte attenuator) antibody is a type of monoclonal antibody designed to target and activate the co-inhibitory receptor BTLA (CD272). BTLA is expressed on various immune cells, including T cells, B cells, and dendritic cells. Upon binding to its ligand, herpesvirus entry mediator (HVEM), BTLA recruits tyrosine phosphatases SHP-1 and SHP-2, which downregulate T-cell receptor (TCR) and B-cell receptor (BCR) signaling pathways. By mimicking this natural interaction, agonistic anti-BTLA antibodies deliver inhibitory signals that suppress the activation, proliferation, and cytokine production of pathogenic T and B cells. This mechanism of action is being investigated for the treatment of autoimmune and inflammatory diseases, such as systemic lupus erythematosus (SLE), psoriasis, atopic dermatitis, and glomerulonephritis, where restoring immune tolerance is therapeutic.
