AGS-003-BLD is an autologous dendritic cell-based immunotherapy developed for the treatment of muscle-invasive bladder cancer. It is a personalized vaccine created by co-electroporating a patient's own dendritic cells with amplified tumor-derived RNA (encoding tumor-specific antigens) and synthetic CD40L RNA. This process is designed to stimulate a robust T-cell mediated immune response against the patient's specific tumor antigens. In clinical studies led by the Mayo Clinic, it has been evaluated as a neoadjuvant therapy in combination with gemcitabine and cisplatin prior to radical cystectomy. The platform was originally developed by Argos Therapeutics, but development for bladder cancer was terminated following the company's financial restructuring and the termination of the pilot clinical trial.
