Overview

AGS-009 is a humanized immunoglobulin G4 (IgG4) monoclonal antibody developed to bind and neutralize most subtypes of human interferon alpha (IFN-α). Its mechanism of action involves inhibiting IFN-α, a cytokine implicated in the pathogenesis of autoimmune diseases such as systemic lupus erythematosus (SLE). By blocking IFN-α, AGS-009 aims to reduce the "interferon signature"—a set of genes upregulated by type I interferons that are often elevated in SLE patients. The drug was administered intravenously and evaluated in phase 1 clinical trials for SLE, where it showed dose-proportional pharmacodynamic effects on gene expression regulated by type I interferons. AGS-009 was initially developed by Argos Therapeutics[1][2][3][4][5][7][8].
